Pantethine, also known as co-enzyme B5, is a highly absorbable and biologically active form of pantothenic acid (vitamin B5). Pantethine is the precursor of co-enzyme A which is essential for carbohydrate, fat and protein metabolism. Co-enzyme A boosts energy levels and enhances physical performance, as it is a vital part of the Krebs energy cycle. Co-enzyme A is also required for the production of red blood cells, sex and adrenal (stress) hormones and brain chemicals (neurotransmitters). Pantethine is a mitochondrial energiser, fighting fatigue and stress. Pantethine also helps support healthy blood fats and reduces cholesterol levels (it lowers LDL [bad cholesterol] and raises HDL [good cholesterol]). Pantethine improves HDL quality and reduces abdominal fat (visceral fat) by transporting fat to the cells of the body, to be used as energy. Pantethine is also important for healthy cardiovascular function through its antioxidant and antiplatelet aggregation activity. Pharmacological Action

Pantethine is a vitamin like dietary supplement. It is the biologically active form of pantothenic acid (Vitamin B5). It is converted to co- enzyme A which is an important part of the Krebs cycle. Pantethine seems to have a beneficial effect on triglyceride and lipoprotein levels by producing cysteamine. The hydrolysis product of cysteamine inhibits acetyl-CoA carboxylase, which in turn reduces triglyceride synthesis. Pantethine might also reduce cholesterol synthesis by inhibiting HMG-CoA reductase, by inhibiting the conversion of lanosterol to cholesterol, and by inhibiting the lipid peroxidation of low-density lipoproteins (LDL). Preliminary research suggests that pantethine might reduce atherosclerosis. Some clinical evidence indicates pantethine might decrease platelet aggregation. Hypotriglyceridemic activity reflects cysteamine-mediated inhibition of the hepatic acetyl-CoA carboxylase, which can be expected to activate hepatic fatty acid oxidation. Hypolipidemic effects appear to be mediated by pantethine's catabolic product cysteamine, a nucleophile that attacks disulfide groups. Pantethine may lower blood acetaldehyde levels following alcohol ingestion. This effect may be partially due to accelerated acetaldehyde oxidation by an interaction between hepatic aldehyde dehydrogenase and pantethine-related intermediates formed in the liver. Panthetine may also play an important role in improving brain function by acting as a substrate in the production of coenzyme A, which promotes the synthesis of acetylcholine, a major neurotransmitter in the brain.

Dosage Directions

Adult and children over 12 years: Take 1 capsule twice daily. Can be taken after meals or on an empty stomach.

Side Effects

Orally, pantethine seems to be well-tolerated. It can cause minor gastrointestinal complaints such as nausea, diarrhoea, and epigastric discomfort.

Contraindications

Surgery: Due to the decreased platelet aggregating affects, pantethine should be discontinued at least 2 weeks before surgery.

Product Interactions

Warfarin and anticoagulants: Can decrease platelet aggregation and thus have an additive effect with anticoagulants including: clopidogrel, warfarin, aspirin and heparin. Herbs: May interact with some herbs and supplements that also affect platelet aggregation. These include the following herbs and supplements include angelica, clove, danshen, fenugreek, feverfew, fish oils, garlic, ginger, horse chestnut, panax ginseng, red clover, turmeric, vitamin E, and others.

Special Precautions

Pantethine might interfere with surgical procedures due to its platelet inhibiting properties. Discontinue at least 2-weeks before planned surgical procedures.
